Target intelligence / Profile preview
Homeobox protein Hox-B7 (HOXB7) is a sequence-specific transcription factor belonging to the Antennapedia (Antp) homeobox family. It contains a conserved homeobox DNA-binding domain and plays a crucial role in the regulation of embryonic development by providing cells with positional identity along the anterior-posterior axis. HOXB7 is also involved in adult tissue processes, especially cell proliferation and differentiation. Overexpression or aberrant expression of HOXB7 is associated with tumor growth, progression, and metastasis in a variety of cancers, where it can influence pathways such as EGFR, FGF2, and TGFβ signaling, and is recognized as a marker of poor prognosis in certain cancer subtypes. The biological effects of HOXB7 may vary between cellular contexts and tumor types, sometimes promoting or counteracting aggressive phenotypes depending on the molecular background[1][2][3][4][5][7].
For drug development, proposed mechanisms include inhibition of HOXB7 expression (e.g., using siRNA or antisense oligonucleotides), disruption of HOXB7 interactions with co-factors like PBX1 or CREB-binding protein, or indirect targeting via downstream signaling pathways such as EGFR or TGFβ signaling[2][4].
