Overview

Homeobox protein Hox-B9 (HOXB9) is a sequence-specific transcription factor belonging to the homeobox (HOX) gene family, located in the HOXB cluster on chromosome 17[1][2]. HOXB9 plays critical roles in embryonic development and adult tissue differentiation and is centrally involved in regulating cell proliferation, differentiation, and developmental patterning[1][2][4]. In cancer, HOXB9 displays context-dependent roles: it can promote tumor proliferation, angiogenesis, invasion, EMT, and metastasis in several cancers (e.g., breast, prostate, lung)[2][5], yet in colon adenocarcinoma, high HOXB9 expression is associated with favorable differentiation, inhibited growth, MET, and better patient outcomes[2]. HOXB9 is also implicated in fibrotic disease, for example by facilitating hypertrophic scar formation through activation of the MAPK signaling pathway and direct interaction with p38 MAPK, affecting extracellular matrix component expression[3]. As a biomarker, HOXB9 can be positive or negative, depending on tumor type, and its modulation represents a potential therapeutic strategy, although no direct drug modulators are currently established in clinical use[2][3][5].
