Homeobox protein Hox-C13 (HOXC13)

Target
HOXC13
Molecular classification
Transcription factor, DNA binding protein
01

Overview

Homeobox protein Hox-C13, encoded by the HOXC13 gene, is a member of the homeobox family of transcription factors that bind DNA in a sequence-specific manner to regulate gene expression. It plays a key role in morphogenesis, specifically in hair follicle and nail development, and controls the expression of hair-specific genes such as FOXQ1. HOXC13 is implicated in the pathogenesis of certain cancers and genetic disorders like ectodermal dysplasia. Mammals possess four homeobox clusters (HOXA, HOXB, HOXC, HOXD), and HOXC13 is located in the HOXC cluster on chromosome 12.

02

Mechanism of action

Not applicable for approved drugs; theoretical mechanisms would include modulation of gene expression pathways (e.g., inhibition of transcriptional activity by small molecules or peptides if a drug existed)

03

Biological functions

Hair follicle differentiationRegulates expression of hair-specific genes (including FOXQ1)MorphogenesisSequence-specific DNA bindingRNA polymerase II-specific transcription activation
04

Disease associations

Cancer (acute myeloid leukemia, melanoma, cervical cancer, oral cavity cancer, hemangioblastoma)Ectodermal dysplasia, hair/nail type
05

Safety considerations

Transcription factors are widely expressed and critical for developmental processes; targeting them can impact a broad range of essential cellular activities, raising concerns for off-target and developmental toxicity
06

Biomarkers

HOXC13 mutations and aberrant expression may act as diagnostic or prognostic biomarkers in cancers and ectodermal dysplasia
