Overview

Homeobox protein Hox-D10 is a sequence-specific transcription factor in the Abd-B homeobox family, encoded by the HOXD10 gene on chromosome 2. It is expressed in developing limb buds and plays a critical role in positional identity during development, limb differentiation, and tissue patterning[1][2][3]. HOXD10 acts as a tumor suppressor that limits cell migration, invasion, and angiogenesis. Its downregulation, often via microRNAs (like miR-10b), is linked to increased tumor invasiveness and metastasis across a range of cancers including glioblastoma, breast, gastric, ovarian, and bladder carcinomas[3]. In addition to oncological associations, mutations or loss of function of HOXD10 can cause congenital malformations such as Wilms’ tumor and congenital vertical talus[1][2][3]. HOXD10 is being investigated as a prognostic biomarker and potential therapeutic target for multiple malignancies, but currently, there are no direct clinical drugs targeting HOXD10[3].
