Overview

Homeobox protein Hox-D4 (HOXD4) is a highly conserved transcription factor belonging to the homeobox gene family, which encodes proteins with a characteristic homeodomain that binds DNA and regulates target gene expression during embryonic development and morphogenesis[1][2][5]. HOXD4 is part of the HOXD gene cluster located on chromosome 2q31-2q37 in humans[1][2]. The protein determines positional values in developing limb buds and is involved in axial patterning, collaborating with other HOX proteins such as HOXB4 to fine-tune gene expression at segmental boundaries, particularly in neurogenesis and hindbrain development[1]. Altered HOXD4 function—including chromatin state changes, microRNA repression, or mutations—can lead to developmental abnormalities (e.g., limb and genital defects) and is implicated in the pathogenesis of various cancers such as lung carcinoma, breast cancer, kidney carcinoma, and some hematological malignancies like acute lymphoblastic leukemia[1]. There are no known therapeutic drugs that directly target HOXD4, and it is not considered a classical therapeutic target, but its expression pattern and regulatory dysfunction are relevant biomarkers in specific cancers and developmental disease contexts[1][2][5].
