Overview

Homeobox protein Hox-D9 (HOXD9) is a highly conserved member of the homeobox family of transcription factors that orchestrates key developmental programs, particularly those governing limb and skeletal morphogenesis in mammals[1][3][4]. It resides on the long arm of chromosome 2 (2q31-2q37) as part of the HOXD cluster[1][3]. HOXD9 regulates gene expression by binding DNA via its homeodomain, and its dosage and epigenetic state have broad effects on cell fate and tissue patterning[3]. Aberrant expression of HOXD9 is implicated in multiple cancers, where it promotes cell proliferation, migration, invasion, cell cycle progression, and suppresses apoptosis, and transcriptionally activates oncogenic targets such as ANGPT2, contributing to tumor progression and chemoresistance[2][3]. Clinically, deletions or mutations affecting HOXD9 or its cluster result in congenital malformations, while elevated HOXD9 levels are associated with poor prognosis and enhanced malignancy in various cancers, highlighting its dual importance in development and disease[1][2][3][4].
