Homeobox protein HOXA10 is a highly conserved transcription factor belonging to the HOX gene family, which plays a fundamental role in embryonic development and body patterning [1, 2]. In adults, it is primarily expressed in the endometrium and bone marrow, where it regulates tissue differentiation, hematopoiesis, and reproductive functions [1, 8]. HOXA10 is a master regulator of endometrial receptivity, with its expression peaking during the window of implantation in response to progesterone and estrogen [1, 6, 12]. Dysregulation of HOXA10, often through epigenetic silencing like DNA hypermethylation, is strongly linked to endometriosis, adenomyosis, and recurrent implantation failure [3, 10, 12]. Conversely, its overexpression or translocation is associated with various malignancies, including acute myeloid leukemia (AML) and gastric cancer [4, 8, 11]. While historically challenging to target directly as a transcription factor, therapeutic strategies focus on modulating its expression through hormonal treatments, vitamin D, or epigenetic modifiers to restore fertility or inhibit tumor progression [3, 9, 10].
HOXA10 acts as a DNA-binding transcription factor that regulates the expression of genes essential for uterine development, endometrial receptivity, and hematopoietic lineage commitment by binding to specific promoter sequences (e.g., 5'-TAAT-3') [1, 8, 15]. It modulates downstream targets such as EMX2, ITGB3 (integrin beta-3), and LIF to coordinate cellular differentiation and implantation [1, 2].
