Overview

Homeobox protein Mohawk (MKX) is a transcription factor belonging to the Iroquois-related (IRX) subclass of homeobox genes, specifically a "three-amino acid loop extension" (TALE) homeodomain protein. MKX is crucial for tendon and ligament development, promoting collagen fiber growth and extracellular matrix production during embryogenesis and in adult tissue homeostasis. MKX represses MyoD and inhibits muscle differentiation, favoring tendon cell fate. Loss of MKX leads to tendon hypoplasia, reduced type I collagen, and abnormal tendon structure in animal models. MKX is also implicated in the maintenance of ligamentous tissues, osteogenic inhibition in specific progenitor cell populations, and as a mechanosensitive regulator of tissue differentiation and repair. MKX is not a classical therapeutic target (e.g., receptor or enzyme), but it is studied as a biomarker and a potential regenerative medicine target for tendon repair and degenerative musculoskeletal diseases.
