Homeobox protein NANOGP8 is a human retrogene and a functional paralog of the canonical pluripotency transcription factor NANOG. While NANOG is well-known for its role in embryonic stem cell self-renewal and maintenance of pluripotency, NANOGP8 is a single-exon, intronless retrogene that arose from NANOG and is uniquely capable among its pseudogene family of encoding a functional protein[2][3][7]. The NANOGP8 protein differs from NANOG by only a few amino acids and exhibits nearly identical reprogramming and DNA-binding activity[2][3]. Unlike NANOG, which is highly expressed in embryonic stem cells, NANOGP8 is predominantly and abundantly expressed in a variety of cancers, including glioblastoma multiforme, where it appears to replace or supplement the role of NANOG as a driver of cancer cell stemness, proliferation, and potentially tumorigenicity[1][2][3][7]. Its expression is often much greater than that of canonical NANOG in cancerous cells, making it a potential cancer stem cell marker and a subject of intense research interest for its fundamental and translational significance[1][2][7]. Due to extensive sequence similarity, accurate molecular distinction between NANOG and NANOGP8 requires careful primer and probe design[2]. Currently, no approved drugs selectively target NANOGP8, but its relevance as an oncogenic driver and cancer stem cell biomarker is increasingly recognized, especially in the context of aggressive brain tumors and other malignancies[1][7].
