Overview

Homeobox protein Nkx-2.2 (NKX2-2) is a homeodomain-containing transcription factor involved in the specification and differentiation of multiple cell lineages during embryonic development and adulthood[1][3][5]. It is critical for oligodendrocyte and neuronal differentiation in the central nervous system, as well as for the proper development and function of pancreatic endocrine cells, especially insulin-producing beta-cells[1][5]. NKX2-2 plays roles in the morphogenesis of the CNS and the pancreas and regulates downstream target genes by acting as a transcriptional repressor, often partnering with corepressor proteins. In pathology, NKX2-2 serves as a highly specific immunohistochemical marker for Ewing sarcoma and may be dysregulated in various cancers, diabetes, and some neurodevelopmental disorders[4][5]. There are no approved drugs that specifically target NKX2-2, and its essential developmental functions make it a challenging therapeutic target.
