Overview

Homeobox protein Nkx-2.3 (NKX2-3) is a transcription factor belonging to the NKX family of homeodomain-containing proteins, which mediates the regulation of gene expression through sequence-specific DNA binding[5][3]. NKX2-3 is essential for development and cell-type specification in multiple tissues, including the small intestine, spleen, and salivary glands[2][4][6]. In mice, knockout of Nkx2-3 results in profound defects in splenic and intestinal morphology, abnormal lymphoid organ development, and altered differentiation of mucous acinar cells in the salivary gland[2][6]. In humans, NKX2-3 has been associated with susceptibility to inflammatory bowel disease and certain lymphomas, particularly marginal-zone B-cell lymphoma, where aberrant expression can drive malignant B-cell signaling and homing by activating B-cell receptor pathways and downstream oncogenic signals such as NF-κB and PI3K-AKT[4][5]. NKX2-3 performs both activating and repressive functions in its target tissues, and its physiological roles are largely restricted to development, differentiation, and homeostasis of specialized cells in the mesenchyme and immune system[2][3][4][6]. No approved drugs or established small molecules specifically target NKX2-3 directly, and it is primarily considered a disease-associated transcriptional regulator rather than a direct therapeutic target[5].
