Homeodomain-interacting protein kinase 1 (HIPK1) is a highly conserved enzyme of the serine/threonine kinase family, predominantly localized in the nucleus. It modulates transcription factor activity—particularly homeodomain-containing proteins—by phosphorylation, acting as both a kinase and co-repressor. HIPK1 participates in cellular stress responses, apoptosis, transcriptional regulation, and cell proliferation. Experimental evidence suggests a role in tumorigenesis, eye and erythroid development, and regulation of organ size. The enzyme is closely related to other HIPK family members (HIPK2, HIPK3, HIPK4) and interacts with targets such as TP53/p53. Functional redundancy and overlapping biological roles are documented among HIPK family kinases[1][3][4][5].
