Overview

Homeodomain-interacting protein kinase 3 (HIPK3) is a serine/threonine-protein kinase belonging to the CMGC kinase family and is closely related to the dual-specificity tyrosine phosphorylation-regulated kinases. HIPK3 is primarily located in the cytosol, nuclear body, and plasma membrane. It participates in the regulation of gene transcription, protein phosphorylation, and negative control of apoptosis through phosphorylation of key substrates such as JUN and RUNX2. HIPK3 also acts as a transcriptional co-regulator for homeodomain-containing transcription factors and steroidogenic genes, modulating pathways involved in cell proliferation, apoptosis, and tissue development. Dysregulation of HIPK3 or its circular RNA (circHIPK3) is implicated in various human diseases, including cancers, cataract, diabetes, neurodegenerative disorders, and fibrotic or proliferative ocular pathologies
