Overview

Homer scaffold protein 1 (HOMER1) is a postsynaptic scaffolding protein expressed primarily in the brain and other tissues, playing an essential role in the assembly and integrity of the postsynaptic density. HOMER1 contains an N-terminal EVH1 (Ena/VASP homology 1) domain, which mediates interaction with proline-rich motifs in several target proteins, including group I metabotropic glutamate receptors (mGluR1, mGluR5), inositol 1,4,5-triphosphate (IP3) receptors, Shank proteins, ryanodine receptors, TRPC channels, and others. The long forms (Homer1b, Homer1c) possess additional C-terminal coiled-coil domains, enabling them to multimerize and cross-link protein complexes, facilitating coupling between surface receptors and intracellular signaling machinery such as calcium release from internal stores. The short form, Homer1a, lacks the coiled-coil domain and acts as a dominant negative regulator, modulating synaptic transmission and contributing to synaptic homeostatic plasticity as an immediate-early gene product induced by neuronal activity. Besides its synaptic roles, HOMER1 also contributes to immune regulation and is implicated in neurodegenerative and psychiatric diseases, but it is not currently considered a direct therapeutic target[1][3][5][6][7].
