Overview

hsa-miR-301a-3p is a mature microRNA that functions as a critical post-transcriptional regulator by binding to the 3-untranslated regions (3-UTRs) of target mRNAs, such as PTEN and IGF1 (NIH, 2022; NIH, 2025). It is frequently upregulated in various cancers, including triple-negative breast cancer and lung squamous cell carcinoma, where it acts as an oncomiR to promote cell proliferation, migration, and invasion while inhibiting apoptosis (NIH, 2024; NIH, 2025). This microRNA exerts its effects primarily through the activation of the PI3K/AKT and NF-kappaB signaling pathways (NIH, 2022; NIH, 2025). Beyond oncology, miR-301a-3p is involved in inflammatory conditions like systemic lupus erythematosus and vascular diseases such as venous ulcers (NIH, 2026; NIH, 2022). Due to its stability in circulating exosomes, it is being investigated as a non-invasive diagnostic and prognostic biomarker, as well as a therapeutic target for antisense oligonucleotide-based interventions (NIH, 2025; NIH, 2026).

Mechanism of action

Binds to the 3-untranslated region (3-UTR) of target mRNAs to induce translational repression or mRNA degradation, thereby modulating signaling pathways such as PI3K/AKT and NF-kappaB (MDPI, 2024; NIH, 2025).
