Overview

hsa-miR-608 is a mature microRNA located within the intronic region of the SREBF2 gene on chromosome 10q24.31 (NIH, 2022). It primarily functions as a tumor suppressor by post-transcriptionally silencing key oncogenic targets such as AKT1, EGFR, and acetylcholinesterase (ACHE) (PubMed, 2022). In various solid tumors, including colorectal, breast, and lung cancer, hsa-miR-608 is frequently downregulated, which contributes to uncontrolled cell proliferation and the evasion of apoptosis (PubMed, 2022). A significant area of research focuses on the single nucleotide polymorphism rs4919510 (C>G) within the mature miR-608 sequence; this variant alters binding affinity to target mRNAs and has been linked to cancer susceptibility, prognosis, and treatment response (PubMed, 2017). Therapeutic development currently explores miR-608 mimics to restore suppressive function in malignant tissues. However, like many RNA-based therapies, targeting hsa-miR-608 faces significant challenges, including the need for stable systemic delivery and the prevention of off-target effects in healthy cells (NIH, 2024).
