Overview

HERPUD2, also known as HERP2, is a membrane protein located predominantly in the endoplasmic reticulum (ER) of human cells and contains a ubiquitin-like (UBL) domain at its N-terminus[1][2]. It is homologous to HERPUD1 (HERP1) and participates in protein quality control by regulating ER-associated degradation (ERAD), assisting in the removal of misfolded or aberrant proteins from the ER[1]. HERPUD2 is constitutively expressed, unlike HERP1, which is strongly upregulated by ER stress[1]. HERPUD2 acts as a scaffold in the HRD1/SEL1L ubiquitin ligase complex, promoting degradation of both glycosylated and non-glycosylated ERAD substrates by recruiting DERL2 to the ERAD machinery; this function is essential for normal protein homeostasis but there is no evidence of direct therapeutic targeting or drug interaction with HERPUD2[1][2][3].
