Homosalate

Molecular classification
Other (salicylate ester; cosmetic ingredient; not a receptor/enzyme/transporter)
01

Overview

Homosalate is an oil-soluble organic compound of the salicylate class, used primarily as a UVB-absorbing agent in sunscreens and cosmetic products. It is not inherently a biological target but acts as an external filter, absorbing ultraviolet B radiation (290–320 nm) and converting the energy to heat, thereby helping to prevent sunburn and reduce the risk of UV-induced skin damage. Despite its utility, it offers only moderate UVB protection and does not shield against UVA rays. Concerns exist regarding its potential for endocrine-disrupting effects and enhancement of dermal absorption of other compounds, resulting in regulatory limitations on its permissible concentrations in products

Other names
Homomenthyl salicylate3,3,5-trimethylcyclohexyl 2-hydroxybenzoateBenzoic acid, 2-hydroxy-, 3,3,5-trimethylcyclohexyl ester
02

Mechanism of action

Absorbs ultraviolet B radiation (290-320 nm) and converts the energy into heat, thereby preventing DNA damage and sunburn in topical formulations

03

Biological functions

Other (UVB light absorption; photoprotection; not involved in signal transduction or cellular processes typical of receptors/enzymes)
04

Disease associations

Other (used to prevent sunburn and reduce skin cancer risk through photoprotection, but not implicated as a disease-associated target)
05

Safety considerations

Potential endocrine-disrupting activity (may weakly interact with estrogen, androgen, and progesterone receptors)Increased absorption of other chemicals through the skin (may enhance penetration of pesticides or other ingredients)Detection in human breast milk after topical use (cause for concern in pregnancy/lactation)Regulatory usage limits due to concerns over systemic exposure (max: 10-15% by region)
