Hook microtubule tethering protein 2 (HOOK2) is a member of the Hook family of microtubule-binding adaptor proteins, primarily localizing at the centrosome throughout the cell cycle[2][5]. HOOK2 links microtubules to subcellular structures and has a critical role in maintaining centrosomal structure and microtubule organization, including the regrowth of microtubules after depolymerization[2][5]. It functions as an adaptor in centrosomal processes by binding to centriolin/CEP110 and interacting with other centrosomal proteins such as PAR6α, aPKC, PAR3, PCM1, Rab8a, and CENP-F[1][2][3]. HOOK2 is an essential component for correct centrosome and Golgi apparatus positioning, polarized cell migration, and the initial steps of ciliogenesis[1][2]. HOOK2 is not established as a receptor, enzyme, transporter, or conventional therapeutic target, and no drugs are currently known to interact with it directly. It does not serve as a common clinical biomarker, nor are there specific safety concerns or therapeutic considerations documented for this molecule[1][2][3][4][5].
