Overview

The host antigen-presenting cell (APC) translational machinery refers to the intracellular system of ribosomes, transfer RNAs, and translation factors within immune cells like dendritic cells that synthesize proteins from mRNA templates (Nature Reviews Drug Discovery, 2018). In the context of mpox (monkeypox) prevention, this machinery is utilized by mRNA-based vaccines, such as Moderna's mRNA-1769, to produce specific viral antigens like A29L, M1R, and B6R within the host's own cells (Moderna, 2024). Once synthesized, these antigens are processed and presented on the cell surface via major histocompatibility complex (MHC) molecules, which is essential for the activation of adaptive B-cell and T-cell responses (Cell Host & Microbe, 2022). While the translational machinery is a fundamental host physiological component rather than a traditional drug target for inhibition, it serves as the critical site of action for nucleic acid therapeutics. This mechanism allows for the induction of a robust and specific immune response against the monkeypox virus without the risks associated with live-attenuated vaccines (The Lancet Infectious Diseases, 2023).
