Overview

Host cell factor C1 regulator 1 (HCFC1R1) is a human protein that modulates the activity of host cell factor C1 (HCFC1), primarily by regulating its subcellular localization[4]. Overexpression of HCFC1R1 leads to the accumulation of HCFC1 in the cytoplasm, and this activity is believed to facilitate the nuclear import of viral proteins such as VP16, a key herpes simplex virus virulence factor[4]. HCFC1R1 is located in the nucleoplasm and is considered an adaptor or regulatory protein, not a classical receptor or enzyme. Although HCFC1 is recognized as a potential therapeutic target in cancer and viral infections, HCFC1R1 itself is not, as of 2025, directly targeted by any drugs, nor is it confirmed as a biomarker for disease. Its biological significance lies in its ability to alter the nuclear-cytoplasmic distribution of HCFC1, potentially influencing cellular gene transcription and viral replication[4].
