Overview

Host cell glycans are carbohydrate moieties covalently linked to proteins and lipids on the plasma membrane, forming the glycocalyx, which mediates numerous cell–cell, host–pathogen, and immune interactions[2][3][5][6][8]. They function as key determinants of viral and microbial attachment, stabilize membrane proteins, protect cells from proteases, and regulate immune recognition and cell signaling[3][5][6][8]. Glycans—such as N-linked or O-linked chains and glycosaminoglycans—are not a single molecule or gene product, but a highly diverse and dynamic collection of structures shaped by the cellular enzymatic repertoire[3][5][8]. As general anchoring factors for pathogens (e.g., heparan sulfate for HIV and coronaviruses), host cell glycans can be targets for antiviral strategies or diagnostic biomarkers, but are not conventional drug targets in the classical sense, since they are complex chemical modifications of numerous proteins and lipids rather than single gene products or receptors[2][3][5][8]. Accordingly, broad mechanistic targeting is challenging and poses substantial safety concerns due to the essential nature of glycans in multicellular physiology[5][6].
