Overview

The host cell-matrix interface is a dynamic and complex structural region where the cell membrane interacts with the extracellular matrix (ECM), providing essential mechanical and biochemical cues (Humphries et al., 2019, Nature Reviews Molecular Cell Biology). This interface is primarily mediated by transmembrane receptors, such as integrins, which link the intracellular cytoskeleton to ECM components like collagen, laminin, and fibronectin (Iskratsch et al., 2014, Nature Reviews Molecular Cell Biology). It serves as a hub for mechanotransduction, allowing cells to convert mechanical stimuli from the ECM into chemical signals that regulate gene expression and cell behavior. Pathological alterations at this interface are central to the progression of cancer metastasis, where tumor cells exploit matrix interactions to migrate, and in fibrotic diseases, where excessive matrix deposition disrupts organ function (Walker et al., 2018, International Journal of Molecular Sciences). Although the interface itself is a multi-component biological structure rather than a single molecular target, many of its constituent proteins, particularly integrins and matrix-degrading enzymes, are significant targets for therapeutic intervention in oncology and inflammatory diseases.
