Overview

The replication factor machinery targeted by the Adeno-associated virus (AAV) Rep78 protein consists of a complex of host cell proteins essential for DNA synthesis and site-specific integration. Rep78, a non-structural viral protein, functions as a site-specific endonuclease and helicase that recognizes the Rep Binding Element (RBE) within the viral inverted terminal repeats (ITRs) and the human AAVS1 locus on chromosome 19 (Dutheil et al., 2000, Journal of Virology). By recruiting host factors such as Proliferating cell nuclear antigen (PCNA), Replication protein A (RPA), and Replication factor C (RFC), Rep78 facilitates the replication of the AAV genome and its integration into the host DNA (Nash et al., 2008, Journal of Virology). Beyond its role in the viral life cycle, Rep78 has been observed to exert potent anti-proliferative effects by interacting with cell cycle regulators like Tumor protein p53 and inducing G1 or G2 phase arrest (Stracker et al., 2004, Journal of Virology). This makes the interaction between Rep78 and the host replication machinery a subject of interest for both gene therapy optimization and potential anti-cancer strategies (UniProt P03132). The machinery is also a critical consideration in the safety profile of AAV vectors, as Rep-mediated effects can influence host genomic stability.
