The host endosomal membrane is the lipid bilayer that defines the endosome, a vesicular organelle in eukaryotic cells critical for sorting, trafficking, and fusion of endocytosed cargo. It contains defining markers (such as specific phosphoinositides like PI3P and protein effectors including Rab GTPases)[1][2], and serves as the site of fusion for many enveloped viruses entering cells via the endocytic pathway[3][4][5]. The makeup and properties of the endosomal membrane determine its identity (early, late, etc.) and influence its fusion specificity, particularly during viral infection or therapeutic delivery. While crucial to intracellular transport and disease mechanisms, the endosomal membrane itself is not considered a classical drug target, but is essential as a platform for molecular interactions and trafficking events.
