Overview

The host epithelial barrier refers to the thin, continuous lining formed by epithelial cells covering mucosal surfaces such as the gastrointestinal tract. These cells—joined by tight junctions and covered by mucus—serve as the frontline physical and biochemical defense against pathogens, toxins, and other antigens. The barrier balances permeability to nutrients and fluids against exclusion of harmful organisms, supporting immune tolerance to commensals while enabling the immune response to pathogens. Dysfunction or disruption of the epithelial barrier is implicated in a broad spectrum of diseases, from chronic inflammation and infection to cancer and metabolic disorders[1][2][3][4][5].
