Overview

Host epithelial cell glycoprotein receptors represent a broad and heterogeneous class of membrane-bound proteins characterized by the presence of oligosaccharide chains covalently attached to their polypeptide side chains. Located predominantly on the apical surface of epithelial cells in the respiratory, gastrointestinal, and urogenital tracts, these molecules are essential for maintaining tissue architecture, mediating cell-cell communication, and providing a protective glycocalyx barrier (PMID: 30634457). Despite their physiological roles, they are frequently exploited by a wide array of pathogens as primary docking sites; for example, influenza viruses utilize sialic acid-containing glycoproteins for attachment, while SARS-CoV-2 targets the glycosylated Angiotensin-Converting Enzyme 2 (ACE2) receptor (PMID: 22230605, PMID: 32132184). In addition to viral entry, these glycoproteins are involved in bacterial adhesion and the inflammatory response, making them significant focal points for infectious disease research (PMID: 11544350). Therapeutic interventions targeting these receptors often involve the use of monoclonal antibodies or small molecules designed to block the interaction between pathogen ligands and the host glycan structures. However, because this term describes a functional category rather than a single molecular entity, it is considered a non-specific target designation in drug discovery contexts.
