Overview

Host gut epithelial proteins represent a heterogeneous and broad category of molecules expressed by the various cell types lining the gastrointestinal tract, including enterocytes, goblet cells, and enteroendocrine cells. This group is not a single therapeutic target but rather a collection of functional and structural proteins such as tight junction components (e.g., claudins, occludin), nutrient transporters (e.g., SGLT1), and signaling receptors like guanylate cyclase-C (GC-C) (Groschwitz & Hogan, 2009, PMID: 19246317). These proteins are fundamental to maintaining the intestinal barrier, which prevents the translocation of pathogens while facilitating the selective absorption of nutrients and ions (Odenwald & Turner, 2017, PMID: 28123271). In clinical practice, specific proteins within this category are targeted to treat gastrointestinal disorders; for instance, GC-C agonists like linaclotide are used for chronic constipation, while GLP-2 analogs like teduglutide promote mucosal growth in short bowel syndrome (Wald, 2016, PMID: 27154402). Dysregulation of these proteins is a primary driver of pathology in inflammatory bowel disease (IBD) and celiac disease, where impaired barrier function leads to chronic immune activation (Lee et al., 2018, PMID: 29467240). Because this term encompasses thousands of distinct proteins with divergent functions, it is considered too broad for specific drug-target classification and typically requires specification of a particular molecular target.
