Overview

Host immune and stromal cell proteins represent a broad and heterogeneous category of molecules produced by the host organism rather than by a pathogen or malignant cell. This group includes cell surface receptors on immune cells, such as PD-1 and CTLA-4, as well as structural and signaling proteins within the stroma, such as collagen, fibroblast activation protein, and vascular endothelial growth factor (Nature Reviews Cancer, 2021). These proteins collectively form the microenvironment that can either suppress or facilitate disease progression, particularly in oncology where they modulate tumor growth, immune evasion, and metastasis (Cell, 2020). Therapeutic strategies targeting these host factors aim to reprogram the local environment to favor an effective immune response or to disrupt the supportive niche provided by stromal cells (Journal of Clinical Investigation, 2017). For example, checkpoint inhibitors target immune cell proteins to restore anti-tumor activity, while anti-angiogenic drugs target host-derived growth factors to starve tumors of nutrients (NEJM, 2018). Because this term encompasses thousands of distinct molecular entities across diverse cell types like T-cells, macrophages, and fibroblasts, it is classified as a functional group or biological compartment rather than a single specific therapeutic target.
