Overview

Host inflammatory pathways encompass the integrated network of molecular signals and cellular processes that mediate an organism's response to harmful stimuli, such as pathogens, damaged cells, or irritants [1, 2]. These pathways are characterized by the activation of pattern recognition receptors, the induction of intracellular signaling hubs like NF-κB and JAK/STAT, and the subsequent release of inflammatory mediators including cytokines, chemokines, and lipid signaling molecules [2, 8, 16]. While these responses are vital for host defense and tissue repair, their dysregulation is a central driver of diverse pathologies, ranging from acute cytokine storms in viral infections to chronic inflammation in autoimmune and metabolic diseases [4, 6, 18]. Therapeutic strategies targeting these pathways, known as host-directed therapies, aim to restore immunological balance by either suppressing excessive inflammation to prevent collateral tissue damage or augmenting specific immune functions to enhance disease resolution [3, 12, 15]. Consequently, drugs such as corticosteroids, cytokine inhibitors, and kinase inhibitors are frequently employed to modulate these pathways across a wide spectrum of clinical indications [4, 9, 13, 17]. Monitoring these pathways often involves measuring systemic biomarkers like C-reactive protein and specific cytokines to assess disease activity and treatment response [5, 7, 10]. However, therapeutic intervention carries risks, primarily related to the potential for over-suppression of the immune system and increased vulnerability to opportunistic infections [3, 11, 17].
