Overview

The Host interferon-related signaling pathways and HIV-1 LTR-dependent transcription machinery represent a complex regulatory network that governs the life cycle and persistence of HIV-1. The HIV-1 Long Terminal Repeat (LTR) functions as the viral promoter, utilizing host transcription factors like NF-κB and the P-TEFb complex to drive the expression of viral genes (Karn & Stoltzfus, 2012). This process is significantly modulated by host interferon (IFN) signaling, primarily through the JAK-STAT pathway, which can induce an antiviral state or contribute to the establishment of viral latency (Doitsh & Greene, 2016). In chronic HIV infection, persistent IFN signaling is associated with immune exhaustion and the maintenance of the latent reservoir. Therapeutic strategies targeting this machinery include the use of JAK inhibitors, such as ruxolitinib, to dampen inflammation and potentially reduce the size of the viral reservoir (Gavegnano et al., 2017). Additionally, latency-reversing agents (LRAs) aim to exploit these transcriptional pathways to shock latent viruses out of hiding for immune clearance. Consequently, this system is a focal point for research into HIV cure strategies, balancing the induction of viral expression with the control of host immune responses. Understanding the interplay between these host pathways and the viral transcription machinery is critical for developing strategies to eradicate the latent HIV reservoir (Bosque & Planelles, 2011).
