Dysregulation of this environment, characterized by a breakdown in barrier integrity or a loss of immunological tolerance to commensal microbes, is a hallmark of diseases such as inflammatory bowel disease, celiac disease, and chronic obstructive pulmonary disease (Neurath, 2014, Nature Reviews Immunology). Pharmacological intervention in this environment typically involves the use of biologics to block specific inflammatory pathways, such as TNF-alpha or integrins, or anti-inflammatory agents to restore homeostasis and promote mucosal healing (Feagan et al., 2013, New England Journal of Medicine).
