Overview

The host nuclear transcription machinery acting on the Chicken Beta-Actin (CBA) promoter is a multi-protein assembly responsible for driving the expression of exogenous genes in gene therapy applications (Miyazaki et al., 1989). This machinery primarily consists of RNA polymerase II and a suite of general transcription factors that recognize the hybrid CBA promoter sequence, which typically includes a CMV enhancer and a chicken beta-actin promoter/intron (Gray et al., 2011). Because the CBA promoter is highly active and constitutive across various mammalian cell types, it is a preferred tool for ensuring robust production of therapeutic proteins in vivo (Addgene, 2023). While not a traditional therapeutic target for small molecules, this transcriptional interaction is the fundamental mechanism by which gene therapies like onasemnogene abeparvovec exert their effect (FDA, 2019). The efficiency of this machinery's recruitment to the promoter determines the level of transgene expression and, consequently, the clinical efficacy of the treatment. Potential challenges include the risk of over-expression leading to cellular stress or the loss of expression over time due to epigenetic silencing of the promoter (Papadakis et al., 2004).
