Overview

The host nuclear transcriptional machinery is a multi-component system comprising RNA polymerase II, general transcription factors, and various regulatory proteins that orchestrate the synthesis of messenger RNA from DNA templates (Cramer, 2019, Nature Reviews Molecular Cell Biology). This machinery is essential for maintaining cellular homeostasis, responding to environmental stimuli, and executing developmental programs. In the context of pathology, many viruses exploit these host proteins to facilitate the transcription of the viral genome, effectively turning the host cell into a virus-producing factory (Wojcechowskyj et al., 2011, PLoS Pathogens). Therapeutic strategies targeting this system, often referred to as host-directed therapies, utilize small molecules to inhibit specific components like Cyclin-dependent kinase 9 (CDK9) or RNA polymerase II itself to suppress viral replication or tumor growth (Krystof et al., 2012, Medicinal Research Reviews). However, because this machinery is vital for the survival of all healthy cells, drugs hitting these targets often face challenges related to high toxicity and a narrow therapeutic index (Bensaude, 2011, Transcription).
