Overview

Host plant proteins (HPPs) are the collective group of endogenous proteins produced by a plant host organism, such as Nicotiana benthamiana, when used as a biofactory for recombinant biopharmaceutical production (Source: Frontiers in Plant Science, 2019). In the context of drug manufacturing, HPPs are classified as process-related impurities rather than therapeutic targets (Source: Plant Biotechnology Journal, 2010). These proteins represent a heterogeneous mixture of enzymes, structural components, and metabolic factors native to the host plant (Source: BioProcess International, 2014). The primary concern regarding residual HPPs in a final drug product is their potential to induce immunogenicity or allergic reactions in patients, as plant-specific sequences or glycans may be recognized as foreign by the human immune system (Source: Current Pharmaceutical Design, 2015). Furthermore, certain host proteases can co-purify with the target biologic, leading to proteolytic degradation and reduced product stability (Source: Frontiers in Plant Science, 2020). Consequently, regulatory agencies require rigorous monitoring and removal of HPPs to ensure the safety, purity, and potency of plant-derived biologics (Source: BioProcess International, 2014). Recent advances in host cell engineering, including CRISPR-mediated gene knockout, aim to reduce the expression of problematic HPPs to streamline downstream processing (Source: Frontiers in Plant Science, 2019).
