The host tissues and immune microenvironment represent the complex, multi-cellular ecosystem surrounding a specific site of interest, such as a tumor or an area of chronic infection. This environment is composed of diverse cell types, including immune cells (T cells, B cells, myeloid-derived suppressor cells), stromal fibroblasts, and endothelial cells, all embedded within a specialized extracellular matrix (Binnewies et al., Nature Medicine, 2018). While not a single molecular target, the microenvironment is the primary context for modern immunotherapies, which aim to shift the balance from a 'cold' (immunosuppressive) to a 'hot' (immunogenic) state. Understanding the spatial and functional dynamics of this environment is critical for predicting drug efficacy and overcoming therapeutic resistance (NIH/NCI, 2023).
