Overview

The host tissues and immune microenvironment represent the complex, multi-cellular ecosystem surrounding a specific site of interest, such as a tumor or an area of chronic infection. This environment is composed of diverse cell types, including immune cells (T cells, B cells, myeloid-derived suppressor cells), stromal fibroblasts, and endothelial cells, all embedded within a specialized extracellular matrix (Binnewies et al., Nature Medicine, 2018). In pathological states like cancer, the microenvironment is often reprogrammed to become immunosuppressive, facilitating disease progression and protecting the lesion from the host's immune system (Hanahan & Weinberg, Cell, 2011). While not a single molecular target, the microenvironment is the primary context for modern immunotherapies, which aim to shift the balance from a 'cold' (immunosuppressive) to a 'hot' (immunogenic) state. Understanding the spatial and functional dynamics of this environment is critical for predicting drug efficacy and overcoming therapeutic resistance (NIH/NCI, 2023).
