Drugs like Omalizumab bind to the Cε3 domain of free IgE, preventing its interaction with high-affinity FcεRI receptors on mast cells and basophils, thereby inhibiting the allergic cascade. Allergen immunotherapy (AIT) works by repeatedly exposing the patient to HDM proteins to induce a shift from an IgE-mediated Th2 response to a regulatory T cell (Treg) response, which promotes the production of 'blocking' IgG4 antibodies that compete with IgE for allergen binding.
