Overview

The HOX protein family consists of homeodomain-containing transcription factors encoded by clustered HOX genes, organized into four main loci (HOX1 on chromosome 7, HOX2 on chromosome 17, HOX3 on chromosome 12, and HOX4 on chromosome 2) in humans, with each locus containing multiple homeoboxes that share sequence homology across groups. These proteins play a fundamental role in embryonic development by regulating anterior-posterior (AP) axis patterning and specifying cell and tissue identity along this axis, a function conserved from flies to vertebrates through interactions with TALE-class cofactors like PBC and Meis proteins via motifs such as the hexapeptide (HX). HOX proteins function as sequence-specific transcription factors, coordinating morphological innovations and body plan formation in bilaterian animals. In disease contexts, particularly cancer, HOX transcription factors serve as biomarkers due to their aberrant expression influencing cell identity and proliferation. While no approved drugs directly target HOX proteins, their clustered genomic organization and evolutionary conservation highlight their potential as therapeutic targets, though challenges arise from their developmental roles and lack of specific small-molecule modulators.
