Overview

HOXA10 antisense RNA (HOXA10-AS), also known as HOXA-AS4, is a long non-coding RNA transcribed from the antisense strand of the HOXA10 gene on chromosome 7q15.2[1][2]. It spans three exons and overlaps with the HOXA10 transcript. HOXA10-AS plays a regulatory role in cancer biology by modulating the expression of HOXA10, a gene critical for embryonic development and a known oncogene in several cancer types[2][6]. Elevated HOXA10-AS expression is strongly associated with increased tumor cell proliferation, survival, EMT, and poor prognosis in diverse malignancies[1][2][6]. Mechanistically, HOXA10-AS acts through RNA-RNA interactions to stabilize HOXA10 transcripts, epigenetic modulation of the HOXA cluster, and scaffolding RNA-protein complexes affecting gene expression[2][4][6]. Recent studies also implicate HOXA10-AS in the regulation of apoptosis, cell cycle progression, and cancer metastasis[1][2]. There are no approved drugs directly targeting HOXA10-AS, but RNA-targeting strategies such as siRNA and antisense oligonucleotides are under preclinical investigation, and HOXA10-AS shows promise as both a diagnostic/prognostic biomarker and a novel therapeutic target in oncology[1][2][3][4][6].
