Overview

HRG and FETUB antisense RNA 1 (HRG-AS1, also known as RP11-573D15.8) is a long non-coding RNA transcribed antisense to HRG and FETUB loci on chromosome 3[1]. It has been shown to regulate proliferation and migration in breast cancer cells; its silencing reduces these malignant phenotypes, indicating an oncogenic role[2]. Expression levels of HRG-AS1 can serve as a negative prognostic biomarker in breast cancer[2]. Additionally, in HIV infection, HRG-AS1 expression is downregulated in elite controllers compared to HIV+ patients, suggesting a role in immune response and potential as a diagnostic biomarker[4]. Mechanistically, HRG-AS1 participates in RNA–RNA and ceRNA networks, potentially modulating important oncogenic and immune-regulatory pathways through interaction with miRNAs and mRNAs such as AKT1, FOXO1, and MAPK3[2][4]. No direct interactions with drugs or evidence for being a classical therapeutic target (receptor, enzyme, etc.) are known.

Mechanism of action

Acts via an RNA–RNA interaction mechanism (as part of competing endogenous RNA/ceRNA networks) to regulate other lncRNAs and influence downstream gene expression miRNA sponge activity affecting gene expression (notably mediated by miRNA MIR6820-001 for some pathways in cancer)

03

Biological functions

Regulation of cell proliferation (notably in cancer cell lines)Regulation of cell migrationModulation of immune microenvironment (associated with T cell and macrophage infiltration in tumors)Potential regulatory interaction with mRNA targets such as AKT1, FOXO1, and MAPK3Negative regulatory correlation with AKT in HIV infection
04

Disease associations

Cancer (notably breast cancer for proliferation/migration, poor prognosis biomarker)Potential diagnostic biomarker in HIV infection and elite controllersOther
05

Biomarkers

Biomarker for breast cancer prognosis (high expression → worse prognosis)Biomarker for distinguishing HIV elite controllers from HIV+ subjects (as part of a biomarker panel)
