Overview

hsa_circ_0067557 is a circular RNA (circRNA) derived from the RBP1 (Retinol Binding Protein 1) gene, located on chromosome 3 (PeerJ). It functions primarily as a non-coding regulatory molecule, often acting as a molecular sponge for microRNAs (miRNAs) or interacting with RNA-binding proteins to modulate gene expression (NIH). In colorectal cancer (CRC), hsa_circ_0067557 is significantly enriched in exosomes secreted by cancer-associated fibroblasts (CAFs) (MDPI). These CAF-derived exosomes transfer the circRNA to CRC cells, where it promotes malignant phenotypes such as increased proliferation, migration, invasion, and resistance to chemotherapeutic agents like 5-fluorouracil (5-FU) by upregulating the oncogenes Lin28A and Lin28B (NIH). Beyond its role in oncology, hsa_circ_0067557 has been implicated in the pathogenesis of preeclampsia, where it is predicted to function within a competing endogenous RNA (ceRNA) network involving miR-1248 and the GNB5 gene to influence the PI3K-Akt signaling pathway (Taylor & Francis). It is also found to be upregulated in gastric cancer-derived mesenchymal stem cells, suggesting a broader role in the tumor microenvironment (Spandidos Publications). Due to its stability in biofluids and its critical role in tumor progression and pregnancy-related disorders, hsa_circ_0067557 is considered a promising biomarker and a potential therapeutic target for RNA-based interventions (NIH).
