Hsp70-binding protein 1 (HSPBP1) is a nucleotide exchange factor and co-chaperone that regulates the activity of Hsp70 family chaperones by modulating ATP-ADP exchange and interfering with ATP binding, thus inhibiting Hsp70 function[2][3]. HSPBP1 plays a key role in regulating protein quality control, inhibiting the ubiquitylation and proteasomal degradation of inducible Hsp70 proteins, and modulating key processes in meiosis, DNA repair, and apoptosis[1][4]. Loss of HSPBP1 impairs proper spermatogenesis and meiosis, resulting in male infertility and a role in the stabilization of proteins like HSPA2 and HSPA1L. In cancer, HSPBP1 has tumor-suppressive properties through interaction with BRCA1 and modulation of DNA repair, with loss of expression promoting genomic instability and resistance to certain therapies[4].
