Overview

HT-29 is a human colorectal adenocarcinoma cell line originally isolated in 1964 from a 44-year-old Caucasian female (ATCC, 2023). It is widely used in oncology research as a model for studying cell differentiation, nutrient transport, and the efficacy of anti-cancer drugs (PubMed, PMID: 1554640). The cell line is characterized by its ability to produce mucus and differentiate into enterocyte-like cells under specific conditions, such as the replacement of glucose with galactose (Wikipedia, 2023). Genetically, HT-29 cells are known to carry mutations in the BRAF (V600E), PIK3CA, and TP53 genes, which are critical for understanding resistance mechanisms in colorectal cancer (COSMIC, 2023). Because HT-29 is a complex biological system consisting of a whole cell, it is classified as a model system rather than a specific therapeutic target (NIH, 2023). It is frequently employed in high-throughput screening to identify potential therapeutic compounds before they move into animal models.
