HT-29 is a well-characterized human colorectal adenocarcinoma cell line established in 1964 from a primary tumor of a 44-year-old Caucasian female (ATCC, 2024; MSKCC, 2024). It is widely utilized in biomedical research as a model for studying colorectal cancer biology, including mechanisms of cell proliferation, differentiation, and apoptosis (Wikipedia, 2024). The cell line is characterized by its epithelial morphology and its ability to differentiate into mature intestinal cells, such as enterocytes or mucus-secreting cells, under specific culture conditions like glucose deprivation (NIH, 2024). Genetically, HT-29 cells carry significant mutations in key oncogenic and tumor suppressor pathways, including TP53 (R273H), BRAF (V600E), and APC, which contribute to their malignant phenotype and resistance to certain therapies (Wikipedia, 2024; ATCC, 2024). While HT-29 is frequently used to screen the efficacy of chemotherapeutic agents like 5-fluorouracil and oxaliplatin, it is a research tool and model system rather than a therapeutic target itself (Cytion, 2024).
