Overview

The hexon protein is the most abundant structural component of the adenovirus capsid, forming the facets of the icosahedral shell. In Human adenovirus type 7 (HAdV-7), the hexon protein is essential for maintaining the physical integrity of the virion and mediating interactions with the host immune system. HAdV-7 is a highly contagious pathogen frequently associated with severe respiratory tract infections, including pneumonia and acute respiratory disease (ARD), particularly in high-density environments. The protein contains seven hypervariable regions (HVRs) that are exposed on the virion surface and serve as the primary targets for neutralizing antibodies produced by the host. Due to its role in infectivity and its high visibility to the immune system, the HAdV-7 hexon protein is a critical target for vaccine development, such as the live oral adenovirus vaccine used by military personnel. Therapeutic approaches include the use of broad-spectrum antivirals that inhibit viral replication and the development of monoclonal antibodies designed to block hexon-mediated entry or facilitate viral clearance.
