Overview

Human betaherpesvirus 5 (HCMV) antigens are a diverse set of proteins encoded by the HCMV genome that play critical roles in viral attachment, entry, replication, and immune evasion [1, 2]. The primary targets for neutralizing antibodies and vaccine development are the surface glycoproteins, most notably glycoprotein B (gB) and the pentameric complex (gH/gL/UL128/UL130/UL131A), which are essential for infecting epithelial and endothelial cells [4]. Other antigens, such as the phosphoprotein 65 (pp65) and immediate-early 1 (IE1) protein, are the dominant targets of the host's T-cell-mediated immune response [2]. HCMV is a significant pathogen in immunocompromised populations, such as transplant recipients and individuals with HIV, and is the leading cause of congenital viral infections worldwide [2]. Therapeutic strategies include small-molecule inhibitors targeting viral enzymes like the DNA polymerase (UL54) or the terminase complex (UL56), as well as monoclonal antibodies and vaccines designed to elicit protective immunity against these viral antigens [3, 5].
