Overview

Human chorionic gonadotropin beta subunit (hCGβ) pre-messenger RNA is the initial transcript produced from the CGB gene cluster, which includes CGB3, CGB5, CGB7, and CGB8 (UniProt: P01233). In normal physiology, the translated protein combines with an alpha subunit to form the heterodimeric hormone hCG, which is essential for maintaining the corpus luteum during pregnancy. However, the beta subunit is frequently overexpressed independently in various non-trophoblastic epithelial cancers, such as those of the bladder, pancreas, and lung (PubMed: 16452204). In these oncogenic contexts, hCGβ acts as an autocrine growth factor that enhances cell survival and provides resistance to apoptosis, often correlating with high-grade tumors and poor patient outcomes (PubMed: 12114458). Therapeutic targeting of the hCGβ pre-mRNA using antisense oligonucleotides (ASOs) or RNA interference (RNAi) aims to deplete the pool of available mRNA, thereby reducing protein synthesis and inhibiting tumor growth. This approach is being explored as a method to sensitize resistant tumors to chemotherapy and provide a targeted treatment for hCGβ-secreting malignancies.
