Overview

CD19 is a transmembrane glycoprotein expressed on all stages of B-lineage cells. It functions as a signaling adaptor molecule and part of the BCR co-receptor complex. Due to its restricted expression pattern, CD19 is a valuable biomarker and therapeutic target in B-cell malignancies. Forced expression in non-B-cell tumors allows for experimental evaluation of CD19-targeted therapies in novel contexts. Resistance mechanisms involve downregulation or loss of CD19 expression, sometimes reversible with demethylating agents.

Mechanism of action

CAR-T therapies bind to CD19 expressing cells and induce cell lysis. Bispecific antibodies bind to both T-cells (via CD3) and CD19 expressing tumor cells, promoting T-cell mediated killing.
