Overview

The Human cytomegalovirus 65 kDa phosphoprotein (pp65, encoded by the UL83 gene) is one of the most abundant structural proteins in HCMV virions, localizing predominantly in the tegument. It is not essential for viral replication in cultured fibroblasts but plays key roles in virion assembly, structural integrity, and the modulation of host immune responses by hindering host interferon regulatory factor 3 (IRF3) activity[1][3][7]. pp65 acts as a dominant target for antiviral T cell responses and is widely used in diagnosis and immune monitoring of HCMV infection. While it does not serve as the direct molecular target for currently approved antiviral drugs, it remains a focus for immunotherapeutic and vaccine development[4][6][7].
