Overview

Human cytomegalovirus (HCMV) immediate-early 2 (IE2) mRNA is a primary transcript of the UL122 gene, playing a pivotal role in the initiation of the viral lytic cycle (PubMed: 12134023). The protein product of this mRNA, IE86, acts as a master transcriptional regulator that transactivates the expression of early and late viral genes while autoregulating its own promoter (UniProt: P06492). Because the IE2 protein is indispensable for viral replication, its mRNA represents a high-value therapeutic target for inhibiting HCMV proliferation. The antisense oligonucleotide drug Fomivirsen was specifically designed to bind to a sequence within the IE2 mRNA, preventing the translation of the IE86 protein and thereby suppressing viral activity (FDA: Vitravene Label). This target is particularly relevant in the context of CMV retinitis, a sight-threatening infection common in severely immunocompromised patients, such as those with advanced HIV/AIDS (NIH: MedlinePlus). While newer systemic therapies have largely superseded antisense injections, IE2 mRNA remains a validated target for sequence-specific antiviral intervention.
